It shouldn't make a difference in terms of accuracy of the adjustment.
Personally, I find it somewhat easier to find the little screw when the lens is set to closest focusing distance, but that's a matter of personal preference.

Dante Stella, several years back, when there was a rangefinder group on Yahoo, wrote about all the problems he had trying to adjust the rangefinder on his Canon 7. Synchronizing the horizontal and vertical rangefinder alignment can apparently be a little difficult. I do recall that he did recommend you set the infinity horizontal adjustment first before trying to set the vertical adjustment. And by setting the infinity adjustment, he meant focusing on something several miles away, not the telephone pole that's across the street.
