Dust spot

Discovered dust spot and line when shooting at F16, using a sensor clear pen and a lot of time I cleared the sensor of the line who where actually grease residue from a previous botch cleaning.
Now I shoot and the line are gone and most of the dust spot, but could not see anything on the sensor even with a loupe.
Checked the lenses and yep dust inside.
Is that the dust spot I'm seeing I'm not to sure ,the lense is about 20 years old.
 
You are very unlikely to see visible dust spots from dust inside a lens. It won't make any difference to the image quality unless there was a lot of dust inside, and then you will only see a loss of contrast.

My guess is that you need to do a wet clean. When a camera is fairly new oil thrown off by the shutter and dust from the mechanism as it beds in can get onto the sensor. Trying to clean it by 'dry' methods only smears the oil around and contaminates things like sticky pads and brushes with the oil. This all settles down over time and cleanings become less frequent and can usually be done with 'dry' methods. I just use an Arctic Butterfly brush now for regular cleans unless I get some sticky dust and then go back to a wet clean.
