This is the second time it has done it: what happens is that I load a tray of film (35mm film, unmounted to be precise), it starts previewing with Vuescan, but at some point the tray stops and the machine lets out a loud, high-pitched tone for a few seconds, after which the tray re-starts but of course it has missed out half a frame.
Anyone have that? It is quite alarming.

I recall experiencing something similar long ago. It happend after Silverfast crashed during scanning. After I restarted SF and tried to rescan the same frame the scanner made a high pitched noise and the holder proceeded all the way out the back. My conclusion at the time was that it must have been a misalignment between scanner and software after the crash.

OK, thanks for that, I suppose I just have to see whether it happens again. It is only the second time - I wondered whether the cogs had engaged properly when I inserted the tray.

If you are on a Mac it could help, to delete the <scanner>.plist.

Why? What should I look for and where?
(I'm not going to be back home until Friday so won't be able to look until then).

I had a similar issue with my minolta scanner.
The unit produced strange noises like those you described.
Deleting the plist(ini) file solved the problem.
Just an idea that could helo you.
You may want to ask a windows user where the ini-files are located.

Make sure you got the latest drivers from the Plustek website installed.
The described behavior occurs in case the scanner looses the correct position of the film holder. It is less likely to occur with the latest drivers.
Otherwise make sure to switch the scanner off and on again so it can reset the position.

I use this thread for my issue too.
I've noticed that when I activate the Multiexposure function the final output results overexposed if compared to the preview or to the output with single exposure... why?
I can't understand this... it turns to get better scans with single exposure... always!

It hasn't happened again - but I do have the latest drivers of course. Just something to keep an eye on I suppose,

I concur. Switching from Minolta 5400 II to 5400 I and vice-versa, I would have this sort of similar issue until I systematically remove the .ini file whenever I switch scanners.
