Yes, the tap can be pried off. There are even tools sold to help with that, but you can probably figure some DIY solution.

Depending on where you live and the quality of the box wine, you and your palate might find it preferable to simply buy new bags. The tap is separate, so you don't even have to bother with prying it off. You fill the bag and then push in the tap (easily done without any tools).

Here in Europe these bags ("bag in box") are available from retailers over Ebay. They are popular among people who are into home made juice, wine and so on. Available in 3 liter, 5 liter or 10 liter size and when you buy ten or twenty packs at a time, the cost is in the penny range per bag. Use them two or three times and then discard them.
