could someone explain the difference between the film set up of the R-d1s. It seems important to understand this set up to properly photograph.
What are the default and how to set them up in order to maximize printing on A3. Thanks.

You might want to just shoot RAW and adjust to your taste in your image editor of choice. Especially given the relatively low MP count of the R-D1, you'll want to preserve as much detail as possible. That's what I do, anyhow.

I recall that the default for "film" is optimized for basic or general all around shooting and three that can be adjusted to suit one's own preferences. As default settings I think they were originally set up for portrait, landscape, and night shooting.

After using the basic setting for a considerable amount of shooting I began experimenting with adjustments and currently find this setting suitable for my visual taste:
Film 1: E=m-, S=M, T=M, C=m-, N=L

Of course ymmv, and I always enjoy learning from others what their preferences are.

And of course shoot RAW.

But what about edge enhance and Noise reduction...
It seems that it will change the pixel nature of the photograph.

Better shoot in Raw and then adjust later in Epson PhotoRaw. The latter also allows you to apply edge enhancement and noise reduction properly, with full control.

Now if you shoot in RAW+JPEG the best is to experiment the film settings. I don't shoot in RAW+JPEG because it will consume more battery/card space. I can always later apply a default setting for RAW to tiff or jpeg bulk conversion and then work on the images I like one by one...
