What is the purpose of the screw in the front cover here? There is nothing behind it.

Tripod mount.

You unscrew it, screw it into the tripod mount on the side, so it doesn't get lost, and you have a tripod mount on the door, like this:

It just keeps the hole clean?

As far as I know, that's pretty much it. It just keeps that hole filled. If the hole goes all the way through, it can keep debris out of the camera body, I suppose, but it rarely goes all the way through. On a lot of the vintage cameras on ebay and elsewhere, that screw is missing.
